Identification of a Brain-specific APC Homologue, APCL, and Its Interaction with /3-Catenin1

چکیده
We isolated a novel gene, !/'(/. that showed significant homology to the adenomatous polyposis coli (APC} tumor suppressor gene. This novel gene, located on chromosome I9pl3.3, encodes a protein of 2303 amino acids that is expressed specifically in the brain. The predicted protein of APCL contains five copies of a 20-amino-acid motif (FXVEXTPXCFSRXSSLSSLS). Like APC, this domain of APCL was able to bind to ß-catenin and deplete the intracellular ß-catenin pool. A reporter-gene assay re vealed that APCL could also regulate interaction of ß-cateninwith T cellspecific transcription factor, although less actively than APC. These re sults suggest that the APCL protein may be involved in the \ViilAVmgless signal pathway, and the identification of a novel relative of APC may provide new insights into the function of APC.

p53-binding Protein 2 and Translocates it to the Perinucleus Adenomatous Polyposis Coli Tumor Suppressor, Binds to APCL, a Central Nervous System-specific Homologue of Updated Version
APCL, a central nervous system-specific sequence homologue of the adenomatous polyposis coli tumor suppressor, can regulate the cytoplasmic level of b-catenin as the adenomatous polyposis coli tumor suppressor does, but its overall biological function remains unclear. Using a yeast two-hybrid system, we attempted to isolate proteins that might associate with the unique COOH-terminus of APCL. Am...

متن کاملDifferent Roles for the Axin Interactions with the SAMP versus the Second Twenty Amino Acid Repeat of Adenomatous Polyposis Coli
Wnt signalling is prevented by the proteosomal degradation of β-catenin, which occurs in a destruction complex containing adenomatous polyposis coli (APC), APC-like (APCL), Axin and Axin2. Truncating mutations of the APC gene result in the constitutive stabilisation of β-catenin and the initiation of colon cancer, although tumour cells tolerate the expression of wild-type APCL. Using the coloca...

Truncating mutations affect the adenomatous polyposis coli (APC) gene in most cases of colon cancer, resulting in the stabilization of β-catenin and uncontrolled cell proliferation. We show here that colon cancer cell lines express also the paralog APC-like (APCL or APC2). RNA interference revealed that it controls the level and/or the activity of β-catenin, but it is less efficient and binds l...

Background and purpose: The mature form of brain-derived neurotrophic factor (BDNF) binds to BDNF/NT-3 growth factors receptor (Trk-B). This binding leads to activation of Ras–MAPK pathway which is integrated with cell growth and proliferation. The BDNF deficiency is correlated with various diseases and affects aging and miscellaneous. In the present study we aimed to design a chimeric LAMP-BDN...
